This antibody recognizes a protein of 39 kDa, which is identified as Aurora kinase B (or Aurora B). Aurora B is a serine/threonine protein kinase and component of the chromosomal passenger complex (CPC), a complex that acts as a key regulator of mitosis. It is important for accurate chromosome segregation, cytokinesis, protein localization to the centromere and kinetochore, correct microtubule-kinetochore attachment, and regulation of the mitotic checkpoint. Aurora B forms a tight complex with inner centrosome protein and survivin. Inactivation of any of these proteins causes similar defects in chromosome segregation. A significant overexpression of Aurora B has been found in a variety of human tumors including non-small cell lung carcinoma, astrocytoma, seminoma and carcinomas of the colon, prostate, endometrium and thyroid. Drebrins (developmentally regulated brain proteins) are cytoplasmic proteins that bind F-actin in the brain and are involved in cell migration, extension of neuronal processes and plasticity of dendrites. There are three isoforms: two embryonic types (E1 and E2); and an adult type (A), generated by alternative RNA splicing from a single Drebrin gene. Drebrins are expressed mainly in brain neurons but are also found in skeletal muscle, heart, placenta, pancreas and kidney. Drebrin has been designated as a marker of the dendritic spine. Decreases in Drebrin levels in the brain have been been associated with Alzheimer's disease and Down syndrome. Recognizes a protein of 40 kDa, identified as CD7 (Workshop IV; Code T165). CD7 is a member of the immunoglobulin gene superfamily. Its N-terminal amino acids 1-107 are highly homologous to Ig kappa-L chains whereas the carboxyl-terminal region of the extracellular domain is proline-rich and has been postulated to form a stalk from which the Ig domain projects. CD7 is expressed on the majority of immature and mature T-lymphocytes, and T cell leukemia. It is also found on natural killer cells, a small subpopulation of normal B cells and on malignant B cells. Cross-linking surface CD7 positively modulates T cell and NK cell activity as measured by calcium fluxes, expression of adhesion molecules, cytokine secretion and proliferation. CD7 associates directly with phosphoinositol 3'-kinase. CD7 ligation induces production of D-3 phosphoinositides and tyrosine phosphorylation.

Recognizes a single protein of 33-34 kDa, identified as the prostate specific antigen (PSA). This MAb is highly specific to PSA and stains prostatic secretory and ductal epithelium in both normal and neoplastic tissues. PSA is a chymotrypsin-like serine protease (kallikrein family) exclusively produced by the prostate epithelium, and abundant in seminal fluid. PSA can be detected in the sera of patients with prostatic carcinoma. It is predominantly complexed to a liver-derived serine protease inhibitor, alpha-1-antichymotrypsin (ACT). A higher proportion of serum PSA is complexed to ACT in prostate cancer than in benign prostate hyperplasia.

The superoxide-generating NADPH oxidase includes a membrane-bound flavocytochrome containing two subunits, gp91-phox and p22-phox, and the cytosolic proteins p47-phox and p67-phox. This antibody recognizes a 32 kDa protein, which is identified as homodimer of galectin-13 (also known as PP13). Galectins are a family of soluble beta-galactoside-binding lectins that modulate cell-to-cell adhesion and cell-to-extracellular matrix (ECM) interactions and play a role in tumor progression, pre-mRNA splicing and apoptosis. One such member, galectin-13, is a 139 amino acid protein that contains one galectin domain. Possessing lysophospholipase activity, galectin-13 exists as a disulfide-linked homodimer. Galectin-13 is suggested to have a developmental role in the placenta and may display immunobiological activity in feto-maternal blood-spaces. Recognizes a cell surface glycoprotein of 80-95 kDa (CD44) on lymphocytes, monocytes, and granulocytes (Leucocyte Typing Workshop V). Its epitope is resistant to digestion by trypsin and chymotrypsin. The CD44 family of glycoproteins exists in a number of variant isoforms, the most common being the standard 85-95 kDa or hematopoietic variant (CD44s). Higher molecular weight isoforms are described in epithelial cells (CD44v), which are believed to function in intercellular adhesion and stromal binding.
